It echoed their fight in round one, in which the Roos struggled through the first half to make a comeback in the second.
But playing catchup to return to a match-winning position tired out the Rovers and robbed them of the four points.
The first quarter was all Picola, with the home side kicking the first four goals of the game.
The seniors felt the loss of playing coach Alex Hay, out with a shoulder injury after just five minutes, denying the side a lot of drive.
The Roos’ first score came at the 24 minute mark when Ray Murphy marked and goaled.
Picola were dominating the centre breaks, putting the Rovers’ defence under constant pressure, although led by James Gilligan at centre half, they held up well.
Quarter time saw Picola lead by 19 points.
The Rovers entered the second quarter positively, with Dylan May booting a goal within the first minute, following a free kick.
Picola responded immediately, following another centre break and followed up with two further goals prior to half time to have a comfortable 32 point lead.
Rovers had several opportunities during the quarter but were unable to convert.
The third quarter saw a much improved effort with Rovers holding Picola scoreless.
James Gilligan played more of an on-ball role and provided plenty of opportunities.
It took 10 minutes to get that first goal of the quarter through Nick Sheldon following a mark, and this was followed up with a running goal to Haydn Mowat just within the 15m.
A courageous mark to Thomas Chandler was converted to put Rovers within a couple of goals behind the opponent.
Another major after the siren to Nick Sheldon following a long and wobbly torpedo gave plenty of momentum going into the final break, with the scores at 7.7 to 6.6.
The last term started with Nick Sheldon goaling at the three minute mark, after having several bounces and running into goal to make the difference 1 point.
A towering mark to John Allitt in the goal square two minutes later meant that Rovers had hit the front for the first time in the match.
Picola steadied and showed more experience to kick the final goal of the match with 10 minutes to goal and held on to win another tight game.
Final score: Picola 9.11 (65) to Rovers 8.8 (56).

In an impressive follow up to their 124 point win in round one, the Rovers’ reserves boasted an incredible 128 domination of Picola United in round two on Saturday.
After a strong start in the first quarter leading by 4 goals at the first break, the second term was more fiercely fought.
The second half opened up for the Rovers kicking 8.3 (51) in the third term and 6.5 (41) in fourth.
The backline was all but impenetrable again, with Picola only able to sneak one goal in the last quarter.
Dylan Mills was once again amongst the best charging out of the backline and clearing the ball with his booming left foot.
Liam Headon played his first game for the club, breaking lines through the midfield and off a half back flank.
Terry Lumbar received a lot of attention from the opposition backman after the show he put on last week, but had plenty of support around him with Matty Horne winding back the clock kicking five majors, Jack Pekin kicking four, and Brock Redden kicking three.
Playing coach John Glowrey congratulated Daniel “Mouse” Mison for playing his 350th club game on the weekend.
